Trabalho de Conclusão de Curso Mineração Genômica de Streptomyces Mad 24 isolado de sedimentos de rios Amazônicos para biocompostos com atividade antimicrobiana antimicroorganismos veterinários.(2022-06-17) Caldas, Luis Felype Garcia de Sousa; Espírito Santo, Edson Francisco do; Espírito Santo, Edson Francisco do; Queiroz, Claudia Afra de; Neves, Kilma Cristiane SilvaBacterial resistance is a problem that affects not only the veterinary clinic, but the entire public health chain, although it, together with other zootechnical areas, plays a very important role in this problem. Currently, new approaches are being carried out to obtain molecules with antibiotic action that can combat multidrug-resistant strains, including genomic mining. It works through bioinformatics tools and presents in silico results of a microorganism or a group of microorganisms. In the veterinary clinic, one of the major problems is bacterial otitis externa in dogs, so isolates from this disease are one of the great candidates for the investigation of new antibiotics, especially when we think that these are commensals of the animal's skin. Therefore, we sought to evaluate the biotechnological potential based on the identification of BGCs (Biosynthetic Gene Clusters) of the Streptomyces Mad 24 isolate aiming at the production of antimicrobial compounds, using bioinformatics tools (in silico) and Screening, Cross- Modified Streak against pathogens isolated from microbiological culture of dogs with otitis externa in BHI (Brain Heart Infusion) medium. When evaluating the genome of a strain of Streptomyces, the presence of 14 clusters associated with the production of antimicrobials was observed, with similarity between 2% and 89%. And the antibacterial screening of the Mad 24 strain against the three isolates, observed an inhibition rate of 76.66%, 52.49% and 19.14%, which demonstrates a great potential of antimicrobial action of Streptomyces Mad 24. However, they are Further studies on the purification and characterization of compounds are necessary for the complete identification of these molecules, considering that the modified Cross-Streak is a screening test to assess whether there is antibiotic potential in the bacterial strain against the isolates.
